Contention 1: In order for a society to thrive as a whole, individuals must prioritize the society?s needs in front of their own. According to Auguste Comte, a French philosopher and the founder of the idea of positivism, the first rule/principle of morality is the supremacy of the regulative needs of society over those self-regarding human instincts that come into play. In other words, place others in front of you. It really doesn?t make much sense concerning the benefit of our own survival, but when a society is stronger, it is able to provide more help and necessary physical or psychological help to those individuals who are in need. This is a small-scale indirect example of reverse psychology, because: 1) If you prioritize society, it will prioritize you 2) You lose something, no matter how small, when you help society?s members in need, whether it be food, money, time, or anything else. 3) After gaining what it needed from you, the member of society will repay you back what you lost if you are in need. Warrant-A direct example and proof of this would be the checks and balance system we use in the US today. Contention 2: Answers the Question: Why should we assume that all of this gaining/repaying is really going to happen? If the issues of the society and its members in need are acted upon by those with the necessary means to solve these issues, naturally they would receive the same treatment due to our innate nature. Innate Nature- the sense of right in wrong naturally existing in one since birth; inborn; native. Innate nature has been proven by scientists to be the ultimate ideology of ethics. a. In a studies done by Jean Henri Fabre and B.F. Skinner on babies and their psychological impacted minds in the late 1920s (proving this to be a withstanding theory), Fabre and Skinner concluded that humans can ?feel? a common, universal, centralizing good when they are born. They all act the same, feel the same, and basically follow this ideal moral standard. They don?t have self-interest; they have a need for survival and inherent feeling of ?good?. b. If we are all born ?good?, then why are some of us bad? Finally, after 3 cycl e extension is be performed at 72C for 5 min to complete any unamplified reaction. Figure 1 shows steps involves in PCR. Development of PCR and identification of DNA as signature molecules for individual leads to introduction of DNA based technique for establishment of parental relation and subsequently for crime and criminal detection. Based on DNA sequence of humans it was found that there are many places in entire DNA that are conserved nucleotide repeats and based on size or length of these sequences they are termed as micro and mini satellite (4). It was found that number of repeat in these sequences varies from person to person and inherited from parents to child makes it ideal choice for criminal identification. Later, this process was termed as DNA fingerprinting. Moreover, development of PCR made this technique more powerful and realistic compared to any other technique for criminal identification, since most of the time, the specimen obtained in the crime site is always in less quantity. This small part of body or body fluids like blood, sperm, saliva or even hair is sufficient to isolate DN A and then amplification with PCR make it possible to do different analysis on it. Here the aim of this experiment was to understand fundamental principle and use of polymerase chain reaction and based on that to understand how PCR is used in DNA fingerprinting based crime investigation and criminal identification 2. Methods: 2.1 Buccal DNA extraction: Ten ml, 0.9% saline solution was rubbed vigorously against the cheeks for 10 seconds. The sample (extract from the bucaal cavity) was then transferred into 15 ml centrifuge tube and centrifuged at 2000 g for 10 minutes for the pellet. Thereafter, 500 l of chelex beads were added into the pellet and resuspended with the chelex by pipetting in and out various times such that there are no visible clumps of cells. Five hundred microliter of the aliquot was transferred into 1.5ml microfuge tube and was boiled into a hot block at 100C for 10 mins. There are approximately 3.5 million children who are not proficient in English language due to their different native language. The United States has received maximum number of new arrivals in 1960s. these arrivals included refugees, illegal and legal immigrants and migrants. As a result, number of enrolled students increased significantly and a large number of new comers were admitted in the public schools of the United States. These newly enrolled students were from different nations so they were proficient in their native languages but their proficiency level regarding English language was very low or none. There were not/hardly able to speak English. To overcome this challenge, several educational policies, and legislation and court decisions were made (Berman et al, 1992). The Bilingual Education Act of 1968, Title VII of the Elementary and Secondary Act was the first step that was taken to overcome the challenge of language barrier. The purpose of this Act was to make sure that all the Mexican American children learn to speak English to an extent that can enable them to actively participate in their educational activities. The role of this Act was later expanded to overcome the language barrier of all non-English speaking children. It is shown in Chavez, (1991, p. 11-12): â€Å"It is not the purpose of the bill to create pockets of different languages through the country †¦ but just to try to make those children fully literate in English†. Several steps were taken to overcome the language barrier. The purpose of all the steps was to enable all non-English speaking children to speak English fluently. An example of such as decision was made in Lau v. Nichols (1974) when the United States Supreme Court declared that every non-English speaking child has got the right to get special assistance in learning English. It stated: There is no equality of treatment merely by providing students with the same facilities, textbooks, teachers, and curriculum; for students who do not understand English are effectively foreclosed from any meaningful education. †¦ Teaching English to the students of Chinese ancestry who do not speak the language is one choice. Giving instruction to the group in Chinese is another. There may be others. (Chavez, 1991, pp. 14-15) A large number of Limited English Proficient students are enrolling in the 10th grade test. It was also found that the number of Hispanic high school graduates was very less all over the country. This was due to large number of students who were dropped out of the school. Although there have been several efforts to improve the English language of non-English speaking students specially Hispanic students but it is found that despite all these efforts, the drop out rate of Hispanic students has not decreased to a significant extent (Hispanic Dropout Project, 1998). Conclusion English language learners are usually exempted from appearing in the assessments that are done for native English speakers. It is quite unfair because non-English speaking students are left behind. It is suggested that non-English speakers should also be allowed to appear in the same assessment programs that are done for English speakers. This will help the non-English speakers to get out of the feeling of left out and they will progress a breast with native English speakers. He developed a very well thought plan and it was terrible idea for the little insult that he had received. He had set the plan up perfectly. He knew what would lure Fortunato in, Montresor used something Fortunato loved to get his revenge. Instead of talking with him, Montresor went his own way, the hard way by planning a murder without being caught. The way the scheme was planned had a lot of thought put into it but Montresor never put a different idea on how he could get over the issue. He assumed murder was the best option but it didnt solve anything it would have created more problems. The vaults are insufferably damp. They are encrusted with nitre. The setting describes southern gothic literature. It is mysterious and violent events had happened. In the beginning of the short story Montresor approaches Fortunato, while Fortunato dressed like a jester which was odd but they were at a carnival. The way Montresor lured him to his house he was able to know no one was home to be suspicious. Montresor took kinda took advantage of Fortunato by knowing his love of wine. Knowing what he likes and cant say no to, was the easy way to get his revenge started and took the matter seriously. It doesnt make sense to why he killed him over the insult. The final moments of conversation between Montresor and Fortunato heighten the horror and suggest that Fortunato ultimately and ironically achieves some type of upper hand over Montresor. Montresor had given Fortunato to turn back before they reached the end because Fortunato had been coughing but he said Enough, the coughrs a mere nothing;it will not kill me. I shall not die of a cough. therefore Fortunato had the chance to save his life he just chose not to take it. In conclusion Montresor took his revenge too far by murdering someone. He could have handled the situation differently by talking to him about of just forget about it. This tells the reader that you can only conclude that in Montresor Family, death is way of punishment for being insulted or threatened. He claims that he has borne a thousand injuries from Fortunato, but he does not go into detail about what these supposed injuries actually are.
